Protein 'jailbreak' helps breast cancer cells live

If the fight against breast cancer were a criminal investigation, then the proteins survivin, HDAC6, CBP, and CRM1 would be among the shadier figures. In that vein, a study to be published in the March 30 Journal of Biological Chemistry is the police report that reveals a key moment for keeping cancer cells alive: survivin's jailbreak from the nucleus, aided and abetted by the other proteins. The research highlights that a protein's location in a cell affects its impact on disease, and offers clear new leads for the investigation.
All four proteins were already under suspicion. Researchers, for example, have already tried to assess what levels of HDAC6 in patients with estrogen-receptor positive breast cancer may mean for their prognosis. The results have been inconclusive. The new research suggests that measuring overall levels may not be enough, said the study's senior author Dr. Rachel Altura, associate professor of pediatrics in The Warren Alpert Medical School of Brown University and a pediatric oncologist at Hasbro Children's Hospital.
"We need to look not only at the levels, but also where is it in the cell," she said.
Altura's emphasis on location comes from what her research team found as they tracked and tweaked the comings and goings of survivin in cells. Inside the nucleus, survivin is no problem. Outside the nucleus, but within the cell, it can prevent normal cell death, allowing cancer cells to persist.
In previous work, Altura and her collaborators established that under normal circumstances, CBP chemically regulates survivin, a process called acetylation, and keeps it in the nucleus. The question in the new work was how survivin gets out.
In a series of experiments, what they observed was that in human and mouse breast cancer cells, HDAC6 gathers at the boundary between the nucleus and the rest of the cell, becomes activated by CBP, then binds survivin and undoes its acetylation. This deacetylation allows survivin to then be shuttled out of the nucleus by CRM1.
In the classic jailbreak, CBP is a corrupt guard who looks the other way as HDAC6, the shovel, is smuggled in. The final accomplice, CRM1, is the tunnel with a getaway car on the other end.
Working the new leads
Altura said the research suggests a clear strategy ? to keep survivin in the nucleus ? and two leads to pursue it, both of which she has already begun working on with collaborators in academia and in the pharmaceutical industry.
One idea is to inhibit HDAC6 in an attempt to prevent it from misregulating the acetylation of survivin. While general HDAC inhibitors are in clinical trials, Altura is optimistic that blocking just HDAC6, using specific inhibitors developed by a colleague in Japan, would have fewer complications.
"You always have to worry about all the things you don't know that you are targeting," she said. "If we can target HDAC6, we can maybe block survivin from coming out of the nucleus and maintain it in its good state."
The other strategy is to block CRM1, Altura said, an idea she is pursuing with a pharmaceutical company in breast cancer cells in the lab. She said preliminary experiments look promising in keeping survivin inside the nucleus and making cancer cells more susceptible to dying.

My Take: I would agree with Cosell here that Tannehill shows more, that means he can excel at the NFL level than Christian Ponder did. I also agree with his take that Tannehill's comfort level in the pocket is even more impressive when you consider that he only started for one-and-a-half seasons. Does that mean Tannehill will be a top 12 pick like Ponder? Of course not. Last year's draft happened during the lockout, so teams faced more uncertainty about how they could address QB situations for the upcoming season. Cleveland is unlikely to spend the No. 4 pick on Tannehill, so it will probably come down to Miami at No. 8 and Kansas City at No. 11.

An international team of scientists, including some from Majorca and the Canary Islands, have described a new type of fossil scops owl, the first extinct bird on the archipelago of Madeira (Portugal). Otus mauli, which was also the first nocturnal bird of prey described in the area, lived on land and became extinct as a result of humans arriving on the island.
Twenty years ago, the German researcher Harald Pieper discovered fossil remains of a small nocturnal bird of prey in Madeira, which, until now, had not been studied in depth. The international team of palaeontologists has shown that the remains belong to a previously unknown extinct species of scops owl, which they have called Otus mauli.
"It has long legs and wings slightly shorter than the continental European scops owl from which it derives" Josep Antoni Alcover, one of the authors of the study and researcher at the Mediterranean Institute for Advanced Studies (IMEDEA), a mixed centre of the university of the Balearic Islands and the Spanish National Research Council (CSIC).
The result of the analysis of the proportions of the remains found, which has been published in the journal Zootaxa, reveals that Otus mauli could be a land inhabiting species that ate invertebrates and "occasionally lizards or birds".
"It is likely that their extinction is linked to the arrival of humans and the fauna they brought with them", Alcover explains. He also points out that their disappearance formed part of a pattern of extinction of the island's species, which occurred in virtually all the islands of the world.
According to researchers, amongst the causes of extinction of this scops owl, the destruction of its habitat is highlighted, as Madeira had a lot of serious fires during the seven years that followed the Portuguese arrival. Furthermore, humans brought new birds with diseases that were unfamiliar to the native species, as well as rats and mice that could prey on eggs of animals that had nests close to the ground.

Golden Gate Equipment Finance said it was retained by a client to source Gulfstream IVs and brokered a 48-month lease with a G-IV owner that will transform a diminishing asset into an income producing asset for both parties involved.
?Golden Gate?s ability to understand our needs was instrumental in getting this third lease done for us,? said the lessee?s CEO. ?We look forward to repeating the process.?
Golden Gate said it is searching for more G-IVs as its client expands flight operations by placing these aircraft with a 14 CFR Part 135 certificate operator.
?Many aircraft once considered assets have lost their missions and have become liabilities,? said John Turnacliff, president of Golden Gate Equipment Finance.
?This lease contract transforms one of those high dollar liabilities back into an income producing asset. While each owner?s financial position can vary greatly, a generic situation such as this could see a swing from a three-quarter million dollar annual expense to a three-quarter million dollar annual profit,? he added.

A condo association in Portsmouth, N.H. is suing a resident for having planted flowers in her yard.
Kimberly Bois has "daisies, bearded irises, lavender, hydrangeas and tulip builbs" around her home in the Atlantic Pointe condominiums, where she has lived since 2008. According to the Portsmouth Herald, many of the plants are heirlooms from her late mother. She told the newspaper that they were planted with the permission of the developer, before the condo board even existed. But the association has sent her 13 certified letters, beginning with a cease-and-desist order and escalating to a demand that she remove the plants and pay thousands of dollars in fines and penalties.
PortsmouthPatch reports that Boise offered to settle the dispute by paying up to $3,000, but that the condo board rejected the offer, instead insisting that she pay all of their legal costs and fines.
"Every month I get a certified letter of what my assessments are, and I'm up to $5,500," Boise told PortsmouthPatch. "It's kind of like they sucked the joy out of owning my first home."
Last month a notice reportedly arrived informing Bois that a lien had been placed on her unit for $4,500 in back fines. The board has also reportedly demanded that she pay the $8,000 in attorney's fees it has accrued in the course of suing her. But Bois is fighting back, with a lawyer of her own: She and a neighbor have hired attorney Paul McEachern.
"I guess it just goes to show that a condominium is the worst form of democracy ever invented," McEachern told the Herald. "They probably have less stringent rules in a penitentiary."
According to McEachern, the condo association wants a judge to order his client to get rid of the flowers, restoring her front yard to its original state -- "crap," in his description. The association also wants the work to be done by board-approved contractors, paid for by Bois.
"It's a goodly amount for a few flowers," McEachern said. "Once you think you've heard everything, you hear something new."
Bois was more blunt in her assessment: "It's the stupidest thing I've encountered in my life."
She and her late mother planted the flowers in mulch beds built by the developer in front of her home. The developer's wife had been planting annuals; Bois offered to plant perennials instead, and says that she has a letter from the developer allowing her to do so. Bois' gardening -- which also included the planting of two evergreen trees and some hydrangea bushes, as well as the watering and weeding of a patch of ground out back -- has been judged "development of the property" by the condo association.
The board contends that it will cost them more to maintain the property with the additional flowers and shrubs. Bois rejects this explanation, suspecting "a vendetta" instead. (She had a previous lawyer send the condo association a letter asking the board to, she says, "leave me alone.")
"It's not a logical response," Complained McEachern, who described himself as a casual gardener and said of the planting: "I was impressed. It looks like it was professionally done."
According to the condo association's attorney, Sandy Roberts, the grounds surrounding the condos are in a common area, subject to the board's control. He says that when Bois was allowed to plant the flowers, she was told the permission was temporary and might be revoked in the future.
"It's not about do you like these flowers or don't you," Roberts told the Herald. "It's a question of was it authorized and is it permanent." He expressed concern that if Bois and her neighbor, who built stone walls and a patio, were allowed to keep their unauthorized landscaping elements, "the guy next door will say, 'I want to do it too.' "
"It's a microcosm of democracy," Roberts said of the condominiums' government. "If you don't have the votes, you don't win."
Of course, this isn't the first time a dispute over homeowners' association bylaws has drawn media scrutiny. Some other recent cases making news:
? Last summer, an HOA filed a cease-and-desist order to stop construction on a house being built for a wounded Army vet in Evans, Ga. The nonprofit group Homes for Our Troops was about to start work on a residence for Amy Sgt. 1st Class Seans Gitens, who was partially paralyzed by an IED in Iraq. According to the HOA, Homes for Our Troops had not submitted the necessary paperwork, a charge the group disputed. An HOA spokesman eventually revealed that some property owners were concerned Gitens' customized, single-floor dwelling would depress home values.
? In Lexington, Ky., the Andover Forest Homeowners' Association demanded that a family remove a therapy playhouse for their 3-year-old son, who suffers from cerebral palsy. Following a public backlash, the HOA relented, temporarily; a state lawmaker said at the time that he planned to introduce legislation making it illegal for an HOA to ban property features necessary for therapy.
? And in Pittsburgh, Pa., an HOA removed a 150-pound Virgin Mary statue from the Vaskos' front yard, saying it would not return it until the family paid fines totaling $4,000. "Would that be extortion or kidnapping?" Steven Vasko. "You tell me, I don't know."

The new iPad went on sale on Friday.As usual, Apple zealots waited in lines at Apple Stores for days to get one, even though Apple now distributes gadgets through lots of other retailers (some of which had no lines at all for the iPad).
And, as usual, Apple carefully and wisely sent free iPads in advance of Friday to a handful of friendly gadget reviewers, all of whom were presumably eager to preserve their royalty status with Apple.
(In a strange inversion of journalistic principles, this sort of bribe is not just tolerated in the gadget-review industry--it bestows status points on the bribe-recipient. So we certainly don't blame Apple for spreading around bribes.)
But all this means that the world has now been treated to a raft of iPad reviews from a contingent of folks who have a strong incentive to view the iPad in a favorable light.
(If you had waited in line for 7 hours for an object you could walk into a store and buy today in 7 minutes, wouldn't you want to persuade yourself that your time hadn't been wasted?)
But now, finally, we'll get more objective views of the new iPad.
For example, on Friday, our gadget guru, Steve Kovach, held two iPads in front of me--the iPad 2 and the new iPad--to show off the new "retina screen" that Apple-approved iPad reviewers are hyperventilating about.
I glanced at the two iPads for a second or two, and, frankly, I couldn't tell the difference. They both looked great. Maybe there is, in fact, a mind-blowing difference that will eventually cause me, too, to hyperventilate, but I couldn't see it a glance. (Of course, I'm still happy with my iPhone 3GS).
The "dictation" feature on the new iPad certainly looks cool, but the downfall of speech-recognition software like this is usually long, complex paragraphs, not simple sentences. So the jury's still out on that.
In a side-by-side comparison of videos shot with the iPad 2 and new iPad, I couldn't tell the difference between the two, at least on a small screen.
So, at least at first glance, some of the most ballyhooed features of the new iPad seemed pretty ho-hum in reality.
Now, if I were in the market for an iPad, I'd certainly spend the extra $100 to get these features. But I'm not in the market for a new iPad, because we already have one (an iPad 1, which still does everything we ask it to do very well).
So, anyway, what do normal folks think of the new iPad?

Hugo Chavez, Venezuela President, Leaving Cuba After Cancer ...
CARACAS, Venezuela ? Venezuelan President Hugo Chavez returned home Friday nearly three weeks after undergoing cancer surgery in Cuba, saying he is praying for life and is confident he will survive and triumph in his re-election bid.
After stepping off the plane, Chavez smiled and waved but also spoke soberly about the path ahead in his struggle against cancer. At times, his expression turned serious.
"I lift up a prayer... in this battle for life," Chavez said in a televised speech on the tarmac. "I'm going to live. We're going to live, and we're going to keep on overcoming. And in that commitment I will give everything, all the spiritual and physical strength that fits in my heart."
Then, he added: "Or rather, that doesn't fit. ... It's a force much bigger than the Caribbean."
Chavez came home as many Venezuelans are wondering about his long-term prospects and about how his health will evolve ahead of the country's Oct. 7 presidential election. Chavez has kept secret some details of his illness, such as the type of cancer, spurring speculation about how his cancer might affect the country's political landscape.
His close allies have assured Venezuelans that there is no Plan B and that Chavez is the only leader of his movement heading into the elections. Chavez was greeted by his vice president and Cabinet ministers, and soldiers standing at attention.
He stepped down the stairs from the plane hand-in-hand with his mother and one of his daughters. He walked gingerly and seemed less energetic than in previous appearances.
At times, Chavez's mother and another of his daughters seemed on the verge of tears. His mother raised a hand to his head to bless him.
Chavez, whose eyes appeared puffy, said he had prayed with his family in the morning before leaving Cuba.
"This new return comes converted into a prayer, a song, a commitment, a prayer to God," Chavez said. "A new prayer of hope in this battle that it's been up to us to fight."
Chavez vowed to win re-election and criticized his political opponents, saying they represent "the agenda of the right."
He accused opposition-aligned Venezuelan news media of trying to provoke fear and uncertainty in the country. "I call on the country to stay calm," he said.
Chavez also said the country's intelligence agencies should be "very attentive to the movements of these groups of the most extremely radical and irrational right." He didn't elaborate on his concerns.
The 57-year-old leader is seeking another six-year term in the October presidential vote. His rival, 39-year-old state governor Henrique Capriles, has criticized Chavez's handling of his cancer, saying that if he were president his health would "be a matter of public knowledge."
Before Chavez's return, his last appearance on Venezuelan television had came Monday night in a video showing him walking with two of his daughters in a garden in Havana. Chavez appeared in prerecorded video footage during his stay in Cuba, but Friday was the first time state television showed live video of him since he left for Havana on Feb. 24.
The president has said his Feb. 26 surgery in Cuba removed a tumor from the same location in the pelvic region where another tumor was removed in June.
After he was diagnosed with cancer last year, Chavez underwent an initial surgery in June that removed a tumor the size of a baseball.
He then had four rounds of chemotherapy and said tests showed no signs of any cancerous cells. But last month, he announced he was returning to Cuba for surgery to have a lesion removed.
Chavez has described the most recent tumor as measuring about 2 centimeters (0.8 inches). He has declined to identify the precise location where the tumors appeared.
He next plans to undergo radiation therapy, although it's unclear how soon that will begin.
"I feel very recovered," Chavez said at the airport. He said he had regained weight and "I'm stepping up the rhythm little by little again."
He also said that as his mentor Fidel Castro saw him off at the airport in Havana, Castro told him: "Chavez, I know you. Tell your people that you have to be disciplined. They're going to understand, and nobody should think that everything has passed already."
"No," Chavez added, "We're overcoming, but we should continue being rigorously disciplined."
Chavez invited supporters to the presidential palace on Saturday, saying he would address them.
The president sought to keep up with government business while in Cuba. Last weekend, his aides were in Havana for a televised meeting where Chavez reviewed projects ranging from subway expansion work to public housing complexes.
Chavez said he's been paying close attention to events in the country. "I haven't stopped being attentive one single day, even on the most difficult ones," he said.
As he got into a Mercedes to leave the airport, his aides crowed around and chanted: "Chavez, we love you!"
